Abstract:

Melphalan as an alkylating agent attaches the alkyl group to nucleotides of DNA. Study on mechanism of thisinteraction by using surface enhanced Raman spectroscopy (SERS) was developed to detect and identifychanges due to alkylating process in Raman Spectra. Here we have applied SERS by using colloidal Agnanoparticles to investigate the interaction of Melphalan with cytosine. After incubating melphalan drug withcytosine for 24 h, at 37˚c some peaks in the SERS spectrum of Cytosine were disappeared and after incubationthe height of the peak, assigned to Amid III group was decreased and this peak slightly shifted to higherwavenumbers. The SERS spectrum of cytosine in aqueous solution has been compared with solid state andresult has been demonstrated that in solid state better signals were obtained than in liquid state.
