Abstract:

The aim of this study was to compare the effect of two oil/water phase mixing methods on solid lipidnanoparicles (SLNs) properties: size, zeta potential and entrapment efficiency. D-α-tocopherol acetate and cephalexinused as lipid and drug model and lipid/drug ratios in 3 levels have been investigated. In method A: all materialsincluding drug, surfactants, water and salt are mixed then added to melted lipid. In method B: drug and surfactants aremixed with lipid, and then water phase added to them. The differential light scattering (DLS) used to measure size andzeta potential of SLNs. Also, high performance liquid chromatography (HPLC) used to define cephalexin entrapmentefficiency. The results showed that the first solubilization method made solid lipid nanoparticles with smaller size andmore anionic zeta potential. In addition, more drug entrapment efficiency achieved by method A .
