Abstract:

High resolution genetic maps are prerequisite for chromosome walking, localization of gene of interests, marker assisted selection, map based cloning and QTL mapping. The population that was used in this research was produced from the F1 hybrid, Fukuho – komugi x oligo – Culm through intergenic crosses between wheat and maize. The number of progeny was 107 in wich 42 was used with the purpose of selecting promising genetic markers for saturating the genetic map. The appreciable merits of AFLP markers (reproducibility and polymorphism,…) makes them appropriate in this study. The average percentage of polymorphism was around 16.6%. In primary data analysis total of 115 AFLP markers were divided in 10 linkage groups and in addition some of the markers remained unlinked. The supplementary data analysis along with SSR markers identified the chromosome loci of the markers. Ultimately, 71.1% of the markers were assigned to genome A , 16.5% genome B and only 3% to genome D. Uneven distribution of markers was related to the clustering of AFLP markers with in heterochromatin regions, especially around the centromere. And it is resulted from the low occurrence of crossing over and the suppression of recombination in these regions.
