Abstract:

Microsatellite loci (also termed short tandem repeats or STRs) on the human Y chromosome are increasingly being used for the study of male population history as well as for forensic analysis and paternity testing. The microsatellite mutation rate id 0/001, and the rate of those on the Y chromosome has been estimated to be 2.1*0/001 per locus in each generation. For this study eight of these wildy used Y chromosome makers Dys425, Dys426, Dys434, Dys435, Dys436, Dys437, Dys438 and Dys439 have been selected in order to analyze more than 1200 males from 21 Iranian ethnics groups. Considering the number of markers employed and the number of samples are to be analyzed, a uniplex PCR system would be every time-consuming and labor intensive.Therefore, we decided to design multiplex PCR systems to enable us to amplify as many as marker possible in just one PCR reaction. In this presentation we will be discussing the stage require for designing multiplex PCR, the creteria should be considered and also some of our results. The multiplex PCR systems we developed are capable to analyze the haplotype diversity among Iranian population without need to purchase the very expensive some commerically available multiplex PCR kits.
