Preparation and evaluation of gelatin nanoparticles by modified coacervation method as drug delivery systems

Abstract:
Natural-Polymerbased nanotechnologies are now proposed as an alternative to classical formulations for drug administration, delivery and targeting. Over the past few decades, there has been considerable interest in developing biodegradable nanoparticle (NPs)as effective drug delivery devices. The possibility of preparing uniform nanoparticles consisting of proteins such as gelatin followed by covalent linkage of avidin was investigated herein. Gelatin nanoparticles were prepared by two-step desolvation. The body distribution of colloidal drug delivery systems was mainly influenced by two physicochemical properties, particle size and surface characteristics. however, characterizing the produced nanoparticles were carried out AFM and SEM. As a colloidal drug delivery system, the essential parameters in fabrication were optimized by Taguchi design method. By introducing 4 factors (temperature, gelatin concentration, agitation speed and the amount of acetone) in 4 levels to the software 16 experiments were carried out and the optimum condition was gained in 50º C, 45 mg/ml gelatin concentration, 80 ml of acetone with 600 rpm agitating had the best result and the nanoparticle size was under 174 nm. The mechanistic of the optimum conditions for preparing protein nanoparticles as well as their characterization are strongly discussed.
