Abstract:

Magnetic nanoparticles are useful for immobilization of enzymes duo to low cost, biocompatibility, non-toxic and etc. Enzymes stability (against the substrate, PH, temperature, etc.) and activity can be increase by immobilization. Plant peroxidases are glycosylation enzymes containing heme and calcium ion. In this study, effects of different concentrations of Fe3O4 nanoparticles were investigated on enzyme activity of Lepidium braba plant peroxidase (LDP). In the first stage, after bacterial culture containing plant peroxidase genes LDP in LB liquid medium, its expression was induced by IPT G. After lysis of the bacteria induced, protein purification was performed using nickel-Sepharose column. Activity of the enzyme was measured in the presence of H2O2 and TMB as substrates in treatment with various concentrations (0-0.9 mM) of Fe3O4 nanoparticles. The results showed that the activity elevated in the presence of various concentrations of nanoparticle by the manner nanoparticles increased. The significance increase of activity was just seen at the presence of the highest nanoparticle concentration
