Abstract:

Cancer is the second cause of death after cardiovascular diseases worldwide. It is known as acomplex disease caused By the interplay of multiple factors involved in its initiation, growth andprogression. . The relationship Between the cancer and the immune system Backs to many yearsparticularly with reference to immune surveillance theory By Bernet in 1957.This conceptualphilosophy of cancer paradigm in this theory has Been categoriTed in three steps, includingEradication, EquiliBrium and Escape. In the first phase, the tumor in a dynamic system isgenerating throughout the life But this is the immune system which recogniTes the tumor anderadicates it. The second phase discusses a period of consistence interaction Between tumor cellsand the immune cells, a co-existence period that no signs and symptoms of cancer are detected. Thefate of the tumor in this phase is depending on the quality and quantity of the host immune keygenes and proteins. In the last phase the chronic pressure of the host immune response on thetumor results in immune selection of a highly replicative and progressive version of tumor cellswhich this may end-up with tumor cells escape and metastasis. Apart from the chemo-radiationtherapy and surgery, advanced cell and gene therapy have provided much hope for the Bettertreatment of cancer. In addition, therapeutical antiBodies have Been flooded to the market somewith high efficacy for the treatment of solid malignancies. The new generation of therapeuticantiBodies has Been designed in a way to Block what we know as the immune check-point inhiBitor.The concept of immune checkpoint inhiBitor in recent years has generated hopes for the treatmentof some uncured cancer cases like some types of lung and melanoma cancers. In this presentation,the concept of regulatory Based target therapy in cancer will Be discussed. The check point controlof the Immune suppression induced By tumor and current immune therapies Based on regulatorymechanisms will Be reviewed.
