Cancer related inflammation was first discovered in nineteenth century. It has been proven that the inflammation aids in cell proliferation, survival of malignant cells, promotes angiogenesis and metastasis, destabilize adaptive immune responses, and alters responses to hormones and chemotherapeutic agents. Thus suppression of tumor related inflammation could be an effective way to prevent tumor growth. Recent studies on Curcumin revealed the anti-inflammatory role of this compound, However, the molecular mechanism is still unknown. Curcumin is the dried root of turmeric and it’s a high value agricultural product which is mainly use as food coloring and flavoring agent. There are several studies showing the role of thrombin in inflammatory diseases. In this study, we investigated the mechanism of anti-inflammatory effect of curcumin on thrombin-induced hyper- permeability in mice skin vessels. We showed that thrombin increases the permeability of skin vessels which is abrogated in the presence of curcumin. Further studies showed that inhibitory effect of curcumin on thrombin signaling is suppressed in the presence of Dorsomorphin, a specific inhibitor of AMPK signaling, suggesting that activation of AMPK signaling by curcumin is essential in anti- inflammatory effect of this compound. These results support curcumin as a novel potential nontoxic herbal anti-inflammatory drug in thrombin-induced inflammation in several diseases including cancer
