Abstract:

Introduction: Gastric cancer is considered as the fourth most common cancer worldwide. MUC 4 is aberrantly expressed in gastric cancer. MUC4 can regulate cell apoptosis via both ErbB2 dependentand independent mechanisms. In this research, we aimed to evaluate the possible association between rs2641726 (C> A) polymorphism of MUC 4 and gastric cancer risk in the Iranian population. Materials& methods: In this case-control study, we enrolled 168 patients vs 66 healthy subjects. Allele-specific primer polymerase chain reaction (ASP-PCR) method was applied to genotype rs2641726 in patients with gastric cancer. Standard cycling was performed under the following conditions: Initial denaturation at 95 ºC for 5 min followed by 35 cycles of 94 ºC for 30 s, 55.6 ºC for 30 s, and 72 ºC for 50 s, and finally 72 ºC for 10 min. Results: According to the hardy Weinberg equilibrium, rs2641726 single nucleotide polymorphism are not associated with the gastric cancer incidence in Iranian population (P> 0.05) but we observed significant association between rs2641726 (C> A) polymorphism and control group (OR = 0.30 for A/C and C/C, P-value =<0.0001 and AIC = 265.9), Metastasis negative (OR = 0.27, P-value= 0.014 and AIC = 195.8), no Smoking (OR = 2.27, P-value =0.0004 and AIC = 92.7), H. pylori positive (OR = 2.05 for C/C, P-value =0.2 and AIC = 175.1) and Stage (Sig (Model Fitting Information) = 0.834). Conclusion: rs2641726 single nucleotide polymorphism of MUC4 gene related to hsa- miR-581 was not associated with gastric cancer incidence but some of histopathological features such as Metastasis negative, H. pylori positive, Stage and no Smoking have significant association with rs2641726 polymorphism
