Abstract:

Obgective: Breast cancer is the most common in women. As drugs currently used lead to undesirable side effects, potentially innocuous phytochemicals are considered as attractive alternatives and are being developed as therapeutic agents. Hesperidin (HT) and hesperidin (HD; HT-7-rutinose) are known flavonoids, reduce the proliferation of many cancer cells. Despite extensive studies on both of them, retinoid effect at the C7 of hesperetin on apoptosis is not still clear. This study compared the effect of HT and HD on caspase-3 activity and survival of breast cancer cell line ,MDA-MB-231, was performed. Method: The cells cultured, after 48 hours treated with various concentrations ofeach reagent hesperetin and hesperidin. after that caspas-3 activity and cell viability was measured by spectrophotometer flouresence. Result: The results of MTT showed that HD in comparison with HT has significant cytotoxic effect on the MDA-MB-231 cells (P<0.05). Sample, which treated with 175 micromolar HD in comparison with the sample that has the same concentration from HT has the most positive effect on caspase-3 enzymatic activity (P<0.05). Conclusion: The results of this study, which was conducted for the first time, demonstrste that rutinoside at C7 in flavonoids may induce apoptosis in MDA-MB-231 cells.
