Abstract:

Introduction: Breast cancer is estrogen-dependent cancers. Cytochrome 1B1 which is one type of cytochrome p450, play key role in the carcinogens of breast and the hormone-dependent tissues. Cytochrome P450 proteins catalyze many reactions, including steroids and other lipids, such as 17-beta-estradiol, and the creation of the role of carcinogens. The 17 beta estradiol (E2) cytochrome able to catechol estrogen metabolite 2-OH-E2 and 4-OH-E2 catalyze that play an important role in breast carcinogenesis. In this study we evaluated the polymorphism rs1056836 from gene CYP1B1 and its correlation to cancer of breast in Tehran. Tumor Samples: This case-control study was conducted on 79 women with breast cancer and 79 healthy women attending Shohadaye Tajrish Hospital in Tehran (February 20014 to March 2015), with a mean age of 48±8 years and 43±6 years, respectively. Genomic DNA was extracted from whole blood by using DNA extraction salting out method. PCR-RFLP amplification and DNA sequencing method was applied to define the genotyping Cytochrome CYP1B1 Val432Leu gene among individuals. Discussion and conclusion: Results suggest that CYP1B1 Val432Leu gene polymorphism may be increased risk of breast cancer. Also set the CYP1B1 gene polymorphisms related to the metabolism of estrogen in breast cancer and other hormone dependent cancers and be used to screen women for breast cancer and also to reduce the cost of treatment.
