The evaluation of cell apoptosis induced by extracts of lemon in canine mammary gland tumors by using flow cytometry

Abstract:

Mammary tumors are the most common tumors in female dogs. According to studies in the field of cancer treatment, day by day the number of chemical compounds for the treatment of cancer (including breast cancer) are used, is added. The cost of some of these chemicals, the lack of appropriate response to these compounds, drug resistance and severe side effects, including factors that need further studies to identify new safe drug (herbal) it requires. In this study was to determine the cytotoxic effect of lemon juice on breast cancer cell lines Dogs (this cell line was cultured from the Pasteur Institute prepared) lemon extract (Citrullus limon) were obtained on percolation method and cultured cell lines in plates of 96 wells and the cells treated with the extract in different concentrations (10, 30, 50, 70, 90, 100, 120) (micrograms per ml) after 48 hours, was evaluated colorimetric method (MTT) and the results were measured using ELISA. In the next step the graph was drawn MTT and the IC50, 98 micrograms per ml was calculated, treatment of cells with the concentration and painted by Annexin-v and PI, with the use of flow cytometry in the efficacy of the herbal compound on the induction of apoptosis, and decreased growth of cancer cells was studied. The results showed a dose-dependent effect of lemon juice on breast tumor cells dog and compare cytotoxic effects at concentrations of 30, 50, 70, 90, 100, 120 to the control group is quite significant (P≤0.05). It should be noted in this study, the cytotoxicity of lemon juice on breast tumor cells in dogs was confirmed for the first time.
