Abstract:
Nanomaterials exhibit significantly mechanical, electronic, magnetic, thermal, catalytic and optical properties in comparison with their bulk counterparts, and have extensively attracted interests. In recent years, there has been an increasing interest in the synthesis of nanosized crystalline metal oxides. The copper oxide nanoparticles(CuO) play important role in industry. This nanoparticle are used widely in Oil, gas, petrochemical and chemical industries. Hence, the effects of CuO on Alkaine phosphatase activities and conformational changes have been investigated by kinetic and fluorescence studies. The results of kinetic studies indicated CuO effected on enzyme activity and increased Vmax. Also obtained results from fluorescence studies showed in presence of CuO fluorescence emission reduced so CuO changed microenvironment of BIALP’s Tryptophan residues and plot decreased with increasing temperature that indicated acted as a quencher. The slope of Stern_Volmer Static quenching occurred which refers to the formation of a ground state fluorophore–quencher complex which cannot emit a photon. Also the sings of calculated thermodynamic parameters (ΔH> 0, ΔS> 0) demonstrated hydrophobic interaction acted as dominant forces in formation of enzyme-quencher complexes
