Abstract:

Vaccines have been in use as a prophylactic strategy for more than 200 years and remain the best defense against the infectious diseases. New formulations should be adopted to improve efficacy of HEV vaccine. Lack of ef-ficient delivery system is a major obstacle for up taking and transporting the antigenic molecules to the cytosol of antigen presenting cells present by immune cells. To this end, HEV candidate vaccine was formulated with archaeosome as biodegradable delivery system.The HEV protein was expressed by prokaryotic expression sys-tem. The protein purified by Ni-NTA affinity chromatography and dialyzed against PBS buffer for overnight. archaeosomes were extracted from methanogenic archi bacteria and synthesized with a certain ratio of HEV protein in PBS. The mean diameter of encapsulated archaeosomes was measured by Zeta sizer instrument. This research aims to make Protein-HEV239-included Nano Archaeosomes that is used for targeted-drug-delivery to cure Hepatitis E and we achieved this goal by loading percentage of 69% and the results of these studies showed that archaeosomes has high ability in loading with recombinant protein.These findings demonstrate that Archaea-somes may be a potent delivery system for HEV candidate vaccine delivery.
