Cytotoxic effect of MSCs treated with purified garlic protein fraction R10b on 4T1 METASTATIC MODEL OF BREAST CANCER

Abstract:

Cancer is the second leading cause of death and morbidity in the world. the regulation of the immune response against cancer cells is indispensable. Recently, there has been increasing evidence that MSCs have the ability to migrate to tumor sites and exert stimulatory or inhibitory effects on cancer cell growth. According to a recent study, the purified garlic protein fraction R10 is composed of 4 distinct subunits. This is the first study on the effects of garlic on MSCs. In this study, Adipose tissue-derived mesenchymal stem cells (AT-MSCs) were isolated and cultured according to standard protocols. Afterwards, AT-MSCs were incubated in DMEM with purified garlic protein fraction R10b. The whole medium was subsequently changed and after incubating MSCs for another 24h in DMEM, the supernatant medium and cells were harvested separately. Eventually, the cytotoxic response of 4T1 cells to AT-MSCs treated with purified garlic protein fraction R10b and their supernatant medium was evaluated using the MTT assay.The statistical analyses of the acquired data revealed:1. R10b fraction had no toxic effect on Adipose-derived mesenchymal stem cells in vitro. 2. R10b fraction by itself had no significant cytotoxic effect on 4T1 tumor cells.3. The supernatant medium of AT-MSCs pre-treated with R10b demonstrated no cytotoxic activity on 4T1 cells.4. AT-MSCs pre-treated with R10b had moderate cytotoxic effects on 4T1 tumor cells.
