Abstract:

Introduction: Breast cancer is the second and the most common cancer among women in the world, but its diagnosis in the early stages increases the chance of recovery and survival of the patient.SFRP1 acts as moderators of the Wnt signaling path and considered as a tumor suppressor gene in various cancer cell lines. Recently the role of Epigenetic alteration in cancer gains huge attraction. In the present study SFRP1 expression was studied in breast cancer tumors compared with normal adjacent also miR-582-3p expression as a regulating micro RNA as well as promoter methylation status of SRRP1 were studied as epigenetic control factors. Materials and Methods: In this project, the Real-Time RT-PCR technique was used to analyze expression of SFRP1 gene and mir-582-3p expression in tumor and normal adjacent breast tissue of 35 patients with ductal carcinoma breast cancer. Also promoter methylation status in genomic DNA was evaluated by MS-PCR. SPSS software and proper statistics methods were used for statistical evaluation.Result: Our data showed down regulation of SFRP1 gene and interestingly upregulation of mir-582-3p and hyper methylation of SFRP1 promoter region as epigenetic mechanisms which control SFRP1 expression in breast tumors compared with normal adjacent tissues.Conclusion: Epigenetic alteration in cancerous tissues may considered as potential cancer related biomarkers.
