Abstract:

Introduction&Aim: Recent studies indicate that of miR-145 plays an important role as tumor suppressor and metastasis in a variety of tumor. This gene can also induce the differentiation of embryonic stem cells. the expression of miR-145 is down-regulated in many cancer sells including breast.KLF4 is necessary transcription factor for maintaining the pluripotency of embryonic stem cells.KLF4 it upregulation in 70 percent of breast tumors. Thus KLF4 is considered as an oncogene. Redusethe of KLF4 self-renewal property cause suppresses the cell migration and invasion of breast cancer cells. The aim of this study is investigation of the effect of miR-145 overexpression on KLF4 gene expression in MDA-MB231 cell line. Method: The gene construct of miR-145 expression vector was designed. then, it was transducted to MDA-MB231 cell line. Total RNA extraction and cDNA synthesized. Gene expression of KLF4 measured by Real-Time PCR and flow cytometry technique and the result were analyzed.Result: Preliminary results indicate the KLF4 downregulation following the overexpression of miR-145 in MDA-MB231 cancer cells, and induces differention so me expect to reduce cancer cells.Conclusion: The results of this study for the first time show, the effect of miR-145 on expression of KLF4 in a breast cancer cell line. This study, according to the theory of cancer stem cells may be effective in the control of the progression of cancer through molecular intervention.
