Abstract:
Introduction.Doxorubicin(DOX) is one of the most commonly used chemotherapy drugs for the early and metastatic breast cancer treatment. One of the challenges in the field of pharmaceutical is the creation of biocompatible, high efficiency and cost-effective carriers for drug delivery systems. Nowadays, molecular researches based on β-cyclodextrin(β-CD) and its derivatives have been broadly studied as an important subject in biochemistry.The β-CD based on water-soluble polymers have been extensively studied and have promising application prospects.Methods.The water-soluble β-CD polymers (β-CDP) were prepared by polymerization ofβ-CD using epichlorohydrin as cross-linking agent under basic conditions. The main purpose of this work was preparation and characterization of the inclusion complex between DOXand β-CDP. The DOX/ β-CDP nanoparticles were prepared by mixing β-CDPand DOX in phosphate-buffered saline at differentweight ratio of DOX to β-CDP at 37 °C for overnight.The cytotoxicity of β-CDP, free DOX and their inclusion complex with various concentrations was evaluated by MTT test(24h-72h) after transfection to the MCF-7 cell line.Results.The obtained polymer contains 49% β-CD in structure and its solubilities in water is 875 g/1000 ml.The achieved complexes were characterized by UV-vis spectroscopy, DLS andzeta potential in the solution whichconfirms the preparation of inclusion complex between the β-CDP and DOX. Conclusion.The results showed that the inhibitory effect of growth and cell proliferation of DOX/ β-CDP is better than the free DOX. Also cell death rate was dependent on the dose of transfected DOX.
