The study of miR-145 overexpression effect on NANOG homeobox protein gene in breast cancer cell line abstract
Introduction & Aim: Evidence suggests that miR-145 have important playes in controlling cellular events and it has inhibitory effect on tumorigenicity and also down-regulated in breast cancer. miR-145 also stimulates the differentiation of stem cells. NANOG is a transcription factor of embryonic stem cells pluripotency. This factor is highly expressed in breast tumor cells and it causes increases metastasis and tumorigenesis increase, also oncogenic function of it has been known. The aim of this study was it investigation of the effect of over-expression of miR-145 on the NANOG gene expression in the MDA-MB231 cell line. Methods: First, construction of expression vector for miR-145 was designed and transduction to the MDA-MB231 cell line was done. Total cellular RNA extracted, cDNA synthesized and NANOG gene expression was measured by Real-Time PCR technique and the results were analyzed.Results: Preliminary data showed down-regulation of nanog gene following miR-145 overexpression in MDA-MB231 cells.Conclusion: The results of this study shows for the first time, the effect of miR-145 on expression of NANOG in a breast cancer cell line. According to cancer stem cells theory, the results of this study maybe useful for controlling of cancer progression in the future investigation.
