Background:Atherosclerosis is a major causes of cardiovascular death worldwide. Myocardial infarction(MI)and ischemic stroke are 2 common comorbidities of this disease. With respect to probability of involvement ofsome particular genes and genetic variants in propensity to both myocardial infarction and ischemic stroke,determination of the most important of risk variants in the population could finally lead to determination of highrisk haplotypes and play a significant role in improvement of prognosis and preventing ischemic complicationsof atherosclerosis and reduce those victims. So, in present study we have evaluated the association of PPAP2B,ZC3HC1 and ADAMTS7 genes polymorphisms with Stroke and Myocardial infarction.Method:Patients were categorized in 4 groups (ischemic stroke,MI patiants, ischemic stroke andmyocardial infarction) in present cross sectional study. Genetic variants associated with thesediseases according to the least amount of P-value were picked up (rs3825807, rs17114036,rs11556924). ARMS PCR method was used. Effects of risk allele of exonic regionpolymorphisms on protein conformation were studied through swiss model and sift softwares.Results: Total number of 360 samples were investigated. Rs11556924 and Rs3825807wereassociated with ischemic stroke and normal allele(T) was significantly more frequent in cases.There was no association between rs17114036 and ischemic stroke, myocardial infarction. Theprediction of single nucleotide changes effects on protein conformation, illustrated that risk alleleof rs3825807(T) caused conformational change.Conclusion:Molecular pathways of atherosclerotic plaque formation in heart and brain overlapeach other in special points. In addition to common pathways, special genes are involved inatherosclerosis in heart and brain.
