Abstract:

Abstract: Regarding the function of BAX gene as a tumor suppressor in the apoptosis process, In the presentstudy, the regulatory area of this gene was hyper methylation in patients with metastatic stomach cancer underchemotherapy. In 30 tissue samples, 30 normal protocols of BAX gene expression were performed by MS PCRtechnique. The results of this study showed a significant relationship between the methylation of this gene andgastric cancer (p <0.05) on tumor and normal samples of these patients. There was also a significant relationshipbetween the BAX gene methylation in the tissue and the TYPE of the cancer, but no relationship was found withother clinical and pathological symptoms of the patients.Introduction: Disturbance in the regulation of DNA methylation patterns in cancer cells leads to an increase inthe level of methylation of the CPG islands, the effective genes in the incidence of cancer. The genes that areeffective in apoptosis are genes that interfere with the methylation pattern of their promoter region, whichreduces the appearance of the gene and the incidence of cancer. Among these genes, BAX tumor suppressor canbe referred to apoptosis pathway. Launching apoptosis in tumor cells is considered a way to treat cancer. This issupported by the fact that the traditional chemical treatments above are triggering apoptosis.Methods: Genomic DNA was extracted using kit from the tissue fixed in paraffin of these 30 patients andnormal individuals. Hyper-methylation study of BAX gene was conducted using PCR special methylation (MSP)method and the results were analyzed through electrophoresis on Agarose gel.Results: Methylation of BAX gene with methylation frequency of 23.3% in tissue has a significant relationshipwith gastric cancer (P˂0.01).There was a significant relationship between the methylation of BAX gene in tissue and tumor TYPE (12,35.3%and P ˂ 0.01). No relationship was found between methylation and grade, stage, note, age, gender and otherpathologic and clinical information of the patients (P> 0.05).Conclusion: There is a relationship between the methylation of BAX gene and gastric carcinoma. Excessivepermeation of BAX gene in gastric cancer patients can be used to detect early disease in first-degree relatives ofpatients. Methylation inhibitors with the potential for drug targeting of BAX can be used as pharmacotherapy.
