DEVELOPING A NOVEL COST-EFFECTIVE CHROMOGENIC CULTURE MEDIUM FOR DETECTION AND ISOLATION OF SALMONELLA SPECIES

Abstract:

Background and Aim:Salmonella species as salmonellosis infection agent invade gastrointestinal tracts leading to gastroenteritis (diarrhea, abdominal cramps, and fever) to enteric fevers (including typhoid fever), which is still a major health problem. Moreover, isolation of Salmonellae from dairy products has caused great destructive impacts on dairy industry. So far various commercial detection media have been proposed for Salmonellae isolation from clinical samples, yet chromogenic culture media led to more specific results with higher recovery rates. But the commercial chromogenic media are very expensive due to application of antibiotics and chromogenic substrates.Methods:Here, we focused on developing a new accurate and efficacious chromogenic media to diagnose and isolate Salmonella strains. Thus we used specific Salmonellae chromogenic substrates, in addition to inhibitors of Proteus, Pseudomonas and Gram-positive strains. Detection of coliforms was also conducted using this method.Results:Though cost-effective, results obtained from observation of clinical samples cultured on the proposed chromogenic medium revealed the high recovery rate of Salmonella species colored in magenta, while coliform strains were isolated in blue-green.Conclusion:Hence, we developed a novel chromogenic medium for Salmonellae, which is also accurate, effective, and cost-effective with rapid identification and high recovery rates for Salmonella and coliform strains in clinical samples.
