Correlation between pathologic finding and mutation in codon 12,13 exon 2 K-RAS gene and codon 1799 Exon 15 B-RAF gene in patient with colorectal cancer of Imam Khomeini Hospital

Abstract:

Introduction : Colorectal Cancer (CRC) is the third most common cancer in the world and forth cause of cancer in Iran. Impact of KRAS and BRAF mutation are essential in target therapy of cancer . the aim of this study is to find out the correlation between pathology report data and KRAS and BRAF mutation by Fast COLD-Real Time PCR and full COLD PCR plus HRM method. Material and Method: Genomic DNA was prepared from total 28 paraffin embedded tissue block with QIAamp DNA FFPE Tissue kit according manufacturer protocol. SYTO9 dye had been used. Finding: Over all KRAS tumor mutation rate in our colorectal cancer (CRC) case was 51.9%, neither of all CRC case harbor BRAF V600E. No meaningful correlation between age and sex parameter and KRAS prevalence were detected. Nevertheless a trend in KRAS mutation rate in mens respect to women (59.3 vs 40.7%) were seen. Prevalence of KRAS mutation in 6th or 7th decade of age was more prominent (66.6%) respect to younger case (less than 50). No meaning ful statistical finding between pathologic report data and mutation rate were detected. Conclusion: Our data has more compatibility with similar report of other researcher. The incidence of CRC has increased with in age over 50. Fast COLDReal Time method in comparison of other method used in Iran are more robust, fast and single tube for KRAS evaluation. Thus with attention to high incidence of KRAS mutation in this study, further evaluation with more case is recommended
