The Antiepileptic Effect of Hespertine-Loaded Nanoparticles in Pentylenetetrazol-Induced Chemical Kindling Model

Abstract:

Epilepsy is one of the most common neurological disorders in the worldwide. Oxidative stress and inflammatory processes play main role in the development of disease. Several studies have shown the anti-inflammatory and anti-oxidant effects of hesperetin in various diseases. However, a major problem of hesperetin application low water solubility. Recently, the use of nanotechnology has been considered as ideal strategy for enhancement of aqueous solubility of hydrophobic drugs. Therefore, we attempt to examine the effect hesperetin-loaded nanoparticles on seizures behavior in animal model of kindling. In this study, NMRI male rats (8-10 weeks) in the range of 25-35 g (animal room of Babol University of Medical Sciences, Iran) were used. Animals were pre-treated by hesperetin or hesperetin nanoparticles 10 days before PTZ injection and their application continued 1h before each PTZ injection. To induce kindling model, PTZ was administrated at dose of 36 mg/kg every other day. Expression levels of TNF-alpha and IL-6 were measured by real time PCR. Our data showed that hesperetin-loaded nanoparticles reduced the seizure behaviors. In addition, the expression levels of TNF- and IL-6 downregulated in animals treated with hesperetin nanoparticles. According to the results hesperetin-loaded nanoparticles might be regarded as a supplementary agent in patients with epilepsy
