General identification and isolation of obligate anaerobic bacteria in patients with periodontitis and its association with genomic polymorphism of TGF_β1 beta using Tetra _ARMs_PCR

Abstract:

Introduction: Periodontitis is a common infectious disease that destroys the tissues that hold the tooth and subsequently loosen and tooth loss. Periodontal diseases are multi-microbial and multifactorial diseases and many factors in the host play a role in determining the susceptibility of people to the disease. The most important anaerobic bacteria involved in the periodontitis infection are: Porphyro-monas gingivialis, Proteella intermedia, Prevotella melanino genica, Fosubacterium nucleatum which can be detected and detected by biochemical methods. TGF-β1 is one of the growth factors and an anti-inflammatory cytokine that plays a decisive role in the repair of periodontal lesions. Considering the clinical importance of this type of infections and its complications, this study was conducted.Materials and Methods: This case-control study was performed by approving the University Ethics Committee and informed written consent in the periodontal department of Tehran Dental School. The sample size was 100 patients, including 50 healthy subjects and 50 patients the periodontal microbial infection was done by sampling. Anaerobic bacteria were identified using anaerobic bacteria culture method (anaerobic gas and jar) and extracted DNA from Tetra_arms_PCR patients‟ blood to determine the association between genomic polymorphism TGF_β1 and anemia Periodontitis is determined.Results: All samples were positive culture. More than 65% of the isolated anaerobic bacteria were obligate: Prevotella, Porphyro-monas, Fosubacterium , Peptostreptococcus anaerobious and results from Tetra_ARMs_PCR. After performing the sequencing and reviewing with SNPP software, the results were observed. Frequency of genotype CC, TT CT was 25, 20 and 5%, and the percentage of control group was 20,24and 6%, the frequency of C and T allele in the patient group was 30 and 70%, and in the control group 64 and 36, there was no significant difference in the frequency of alleles and genotypes between the two groups.Conclusion: there is a significant relationship between polymorphism There is no genotype TGF_β1 and periodontitis.
