Abstract:

Background and Aim: Beige/Brown Adipose Tissue (BBAT) – a hyper-metabolic tissue in human body - has been a promising tool to combat metabolic diseases such as obesity, diabetes and metabolic syndrome. Stem cells are able to self-renew and capable of differentiation, or specialization into lineage-specific cell type. The in vitro differentiation of ADSCs into multiple cell types of mesodermal origin has been shown in a variety of studies. Some studies have described the plasticity of ADSCs towards adipocytes. By considering some experimental studies, we aimed to examine the human adipose stem cell potential to brown adipocyte.Methods: To induce adipogenesis, human adipose stem cells were harvested in passage 4 and re-plated in six-well plates. Cells treated with dexamethasone, IBMX, rosiglitazone and insulin during differentiation. After 18 days oil red o staining was performed to confirm the successful adipogenic differentiation.Results: after 18 days of culture, formalin-fixed cells stained with Oil Red and lipid droplets were observed by a light microscope. Oil red staining was representative of oil droplets in differentiated adipocytes.Conclusion: Overall, our study demonstrated that human adipose stem cell can be differentiate to brown adipocyte in differentiation induction cocktail
