Preparation of chitosan /fibrinogen hydrogel-loaded nisin for skin wounds healing

Abstract:
The present study was aimed to design an antibacterial and biodegradable composite hydrogel for skin wound healing. The composite hydrogel was fabricated by emulsion crosslinking method. Fibrinogen plays an important role in anticoagulant control in controlling bleeding and accelerating the process of clotting and repairing and restoring the skin at the wound site. Chitosan possess antibacterial activity against gram-negative bacteria. In order to enhance antibacterial and mechanical properties of prepared hydrogel, nicin loaded chitosan /fibrinogen hydrogel to generate a composite gel dressing. Nicin acts also against gram- positive bacteria. Therefore, synthesized hydrogel have antibacterial effects on the both gram-positive and gram-negative bacteria. The mechanical properties of prepared hydrogel such as swelling, degradation, compressive modulus and rheological properties of the gel dressing were evaluated. The mechanical properties of this hydrogel in dry and swollen state were importantly improved with high swelling ratio. Also, in vitro drug release profiles demonstrated that the loaded nicin could be sustained release from the composite gel dressing by contrast with pure hydrogels. Considering to powerful bacteria growth inhibition effects of produced hydrogel against Escherichia coli and Staphylococcus aureus, chitosan /fibrinogen hydrogel- loaded nisin are potentially suggested for treatment of bacterial infection after further required evaluations.
