Efficient chemical defined differentiation of the definitive endoderm from human adipose derived mesenchymal stem cells

Abstract:

Administration of human adipose derived mesenchymal stem cells (ADMSCs) has recentlyconsidered as a new therapeutic approach in clinical trials for widespread indications includingneurological, cardiac, haematological and pulmonary disorders. However efforts to exploit thesecells have been met with only modest success mainly due to the low efficiency of differentiationand expansion of large quantities of desired cell derivatives under good manufacturing practice. In present study, we focused on the application of various small molecules to differentiatehuman ADMSCs into the definitive endoderm lineage as the initial step in differentiation intoother endoderm derivatives, such as lung, liver, and gastrointestinal tract cells. We found achemical compound which induced nearly 90% of ADMSCs to form definitive endoderm, ahigher efficiency than that achieved with commonly used protein or combinational chemicalinducers of endoderm. These finding can led to improve current approach toward achieving areproducible and efficient differentiation, verification, and application of the ADMSCs for clinicalpurposes
