Abstract:
The aim of this study was investigation the influence of gelatin on cell proliferation of PCL based nanofibers for skin tissue engineering and wound dressing applications. Electrospinning was performed using PCL and PCL/Type B gelatin. PCL solution was prepared in chloroform/dimethylformamid ratio of 2:1 (oil phase) and gelatin solutions was prepared in acetic acid (aqueous phase). The surfactant was added into the aqueous solution then oil phase was added to the aqueous phase at room temperature and stirred for 12 h to obtain a homogenous mixture. Electrospinning was carried out using 27 KV applied voltage, 1 ml/h feed rate, and 17 cm working distance. Fibroblast cell morphology, attachment and proliferation on PCL and PCL/type B gelatin nanofibers after 1, 7, 14 days were observed under Rhodamine-DAPI staining. The DAPI micrographs of HDF cells on the nanofibrous structures show highly attachment and proliferation on the PCL/Type B gelatin nanofibers surface. The PCL/ Type B gelatin electrospun nanofibrous mats have a great potential for using as a scaffold for skin tissue engineering and wound dressing
