Abstract:

Recent advances in chromatin biology have revealed a central role of transcriptional enhancers in regulating the spatiotempo-ral gene expression in mammalian cells. These regulatory ele-ments interact with promoters and other enhancers to generate a complex and cell-specific network of chromatin interactions. This interaction network is crucial for proper regulation of gene expression and 3D genome organization. How enhancers are regulated during early embryonic development, however, is not well understood. In this presentation, I will focus on em-bryonic stem cells and discuss how the enhancer landscape is reprogrammed in different states of pluripotency. Using data obtained from different genome wide technologies, I will ex-amine the mechanisms underlying enhancer activation and the extent to which enhancer-promoter rewiring contributes to spa-tiotemporal gene expression in different states of embryonic stem cells.
