Evaluation of The Effect of Ketorolac on The Rate of Sperm Motility and Survival in Infertile Men with Oligo-Asthenospermia in Culture

Abstract:

Background: Decreased semen quality is a major factor of male infertility. The primary mechanism of action responsible for ketorolac s anti-inflammatory, antipyretic and analgesic ef-fects is the inhibition of prostaglandin synthesis by competitive blocking of the enzyme cyclooxygenase (COX) ۱ and ۲. It has been hypothesized that Ketrolac has impact on semen quality and quantity. In this work, we review and reveal the effect of ketorolac on sperm motility and vitality in infertile men suffer-ing from oligoasthenoteratozoospermia in vitro.Materials and Methods: In this study, ۴۰ seminal fluids were obtained from ۴۰ infertile males suffering from oligoasthe-noteratozoospermia. After collection by floating method, the samples were randomly placed in ISM۱ media in four differ-ent groups: the control group without any addition, group num-ber one with ۵۰ug/ml ketorolc, group number two with ۱۰۰ug/ ml ketorolc, and group number three with ۲۰۰ug/ml ketorolc added to the media. Sperm motility and vitality were examined after ۱, ۲۴, ۳۶, and ۴۸ hours. The data was analyzed by SPSS software. A P value of less than ۰.۰۵ was considered statisti-cally significant.Results: The average percentage of motile sperms was meas-ured in control group after the intervals of ۱, ۲۴, ۳۶, and ۴۸ hours and a significant reduction of motility (P<۰.۰۰۱) was ob-served in higher time points compared to the first hour; there was no statistically significant difference (P=۱۹.۰) between the four groups. The average percentage of alive sperms was also measured in control group after the intervals of ۱, ۲۴, ۳۶, and ۴۸ hours and a significant reduction of vitality (P<۰.۰۰۱) was observed in higher time points compared to the first hour; there was a statistically significant higher decrease in the third group (P=۰.۰۱).Conclusion: Ketrolac increases the sperm vitality in infertile males suffering from Oligo-Asthenospermia only in ۲۰۰ug/ml dosage but has no effect on sperm motility.
