Abstract:
Sometimes extraction of DNA from halophyte plant tissues is difficult. Usually, the extracted DNA is low or low quality and PCR does not produce the desired band. The use of extraction kits from different companies is sometimes inadequate and offers different amounts of DNA and quality, which is not suitable for subsequent steps such as PCR.After several experiments, we optimized the extraction procedure of Thomson and Murray and finally extracted the desired quality and quantity from fresh leaf samples, mature DNA plant leaves, Carpobrotu rossii. DNA concentration was determined by spectrophotometry as well as the color quality of the specimens. It was also colorless and transparent and at the right concentration. But the extracted DNA by the Brown Murray and Thomson method was unrecordable with a lower concentration than tho optimized method.
