Background and Aim : Cervical-cancer is among the most commonly diagnosed cancer in woman, and the presence of HPV is reported to be related with increased inflammatory cytokines. Here we explored the association between a genetic variant in Interleukin-10 (IL-10) and cervical-cancer-risk and its relationship with IFN-γ, TGF-β, TNF-α in 350 cases with/without cervical-cancer. Methods : This case-control study comprised 146 patients with cervical-cancer and 169 healthy controls. Serum IFN-γ, TGF-β and TNF-α levels were measured, while Taq-man®‐probes‐based assay was used to determine genotypes of the polymorphism. Results : Our data showed a significant association between the AA-genotype and the presence of cervical-cancer. In particular the regression models in recessive model, showed that patients with AG/AA genotypes had a higher risk of developing cervical cancer with odds ratio (OR) of 2.85 (95% confidence interval [CI]: 1.07-7.56, p=0.03), compared with the GG genotype. Also the IFN-γ, TGF-β and TNF-α levels in cervix tumor tissue were significantly higher in patients (P<0.001) and were associated with AA genotype. Conclusion : our finding demonstrated the association of the gene variant in IL-10 gene with risk of developing cervical-cancer and higher level of inflammatory markers, indicating further investigations on the value of emerging marker for risk stratification in management of cervical cancer patients.
