Abstract:

The CRISPR/Cas9 system is a powerful gene/genome editing tool in eukaryotes providing new platforms for gene function analysis, human disease modeling, and gene therapy. The capacity of embryonic stem cells to rapidly proliferate and differentiate into numerous specialized cells & tissues, makes them a highly useful device in exploring human physiology and disease. Regarding our recent work on targeting PRKDC gene (player in recombination and repair system) in mouse embryonic stem cells by employing CRISPR/as9, here we discuss the fundamentals of PRKDC knockout modeling, its effect on stemness and development, potential challenges in applying method and, finally future directions
