Abstract:

Background: Wound healing and reducing its recovery time is one of the most important aspects of medical and surgical science. One of the goals of medical treatment in wound healing is shorter and with fewer side effects. That is why this study is to investigate the synergic effect of topical application of nano liposomes containing curcumin and aloe vera gel on the healing of open skin wound bed rats, design and results of this study show that the use of Nanoliposome of curcumin and aloe vera gel as significant in wound healing They re effective.Methods: This experimental study was performed on 63 healthy wistar male rats weighing 300-250 g and aged 3 to 4 months. On the test day under standard conditions, after anesthesia of the rats, open skin ulcers with area include 4 centimeters square were made by bistory blade. The rats were randomly divided into 7 groups of 9. The day of the experiment was considered zero, the control group did not receive any drugs, and for the vehicle group, 1% phenytoin ointment was applied topically. In other groups, the culture medium and adipose tissue stem cells were injected throughout the course of the day and the nanoliposome of curcumin was applied topically. In this research, stem cells were removed from the adipose tissue that situated on the upper border of epididymes in abdominal wall and the stem cell of fatty tissue was confirmed by the flowcytometry. Fusion method was used to prepare the liposome and confirmation of nano-liposomal particles was done by using DLS microscope. In this experiment, 36 rats were divided into 4 groups, and the days were 4, 7, and 14 days. The surface area of the wounds and the percentage of recovery every day was measured with IMAGE J, but other measurements were only reviewed on days 4, 7 and 14. For gene reviews, RT-PCR technique was used to count the fibroblasts from immuno-histochemistry and to measure and count the cells from IMAGE J, and to evaluate the SSPS-edu 22, one-way ANOVA and TUKEY. Tri-chromosome staining was used to determine the collagen level and H & E staining was used to count the number of capillaries.Results: The results of the experiments showed that the TGF-β1 gene expression, vascular number, wound healing rate and fibroblasts increased significantly in the primary treatment group containing adipose stem cell and other group containing nanoliposome of curcumin and the wound surface was also decreased Has meaning. According to the results of this study, it can be said that adipose stem cell and nanoliposome of curcumin can wound healing by rather speed specially in adipose stem cell group than other materials.so it must be said that this material can use to wound healing in the future .
