Correlation of Foxo1 and hsa-miR490-5p expression in peripheral blood mononuclear cells of patients with breast cancer

Abstract:
Introduction: T Regulatory cells (T-reg) are one of the effective immune cells that play an important role in cancers, autoimmune diseases, and infectious diseases. A transcription factor, Foxp3 plays a key role in T-reg cell development and function. However, its balance with FOXO1 can be effective in the fate of T-reg cells. There were several reports about the role of miRNAs in regulating of T-reg differentiation via targeting the FOXO1. One of these microRNAs is hsa-miR-490-5p. One of the anticipated targets of hsa-miR-490-5p is Foxo1. The aim of current study is evaluation the correlation of Foxo1 and hsa-miR490-5p expression in PBMCs of patients with breast cancer.Materials and methods: Peripheral blood samples were collected from 40 breast cancer patients and 40 healthy individuals as a control group. Then, PBMCs of samples were isolated by Ficoll gradient. Next, RNA extracted and cDNA synthesized for each sample. Finally, the expression changes of both miR-490-5p and FOXO1 were evaluated by real time PCR technique.Results: The results of current study showed dramatically over expression in miR-490-5p (25 folds) and a significant reduction in FOXO1 (3.5 folds) levels. It must be added that statistical analysis revealed a negative and significant correlation between miR-490-5p increasing and FOXO1 decreasing.Conclusion: the results showed the potential of miR-490-5p in targeting of FOXO1, and its probable role in T-reg differentiation in the patients with breast cancer.
