In order to optimize tomato (Solanum Lycopersicum) in vitro regeneration, the effects of three different types of plant growth regulators on the regeneration of tomato explants were evaluated. Three different experiments were analyzed. First the effect of five concentrations 0, 0.5, 1, 1.5 and 2 mg/L of Zeatin on regeneration percentage of two types of explants derived from hypocotyls and cotyledons; and two cultivars (Urbana and Cal J) was assessed as a factorial design in a completely randomized design with three replications. In the second and third experiments, Naphthaleneacetic acid (NAA) (0, 0.1, 0.3 and 0.5 mg/L) and Indole-3-acetic acid (IAA) (0, 0.5, 1 and 1.5 mg/L) along with 1 mg/L Zeatin as a fixed factor were analyzed. Results showed that the regeneration in all of the explants increased significantly with the presence of Zeatin. The best regeneration for hypocotyl was achieved on the media with 1 mg/L Zeatin (100%), whereas it was obtained with 2 mg/L Zeatin for cotyledons. The best result for the second and third experiments was recorded on the media with 0.3 mg/L NAA for cotyledons of Cal J (100%) and the media with 1mg/L IAA on the cotyledons for both of the cultivars (81%). Totally in this study we showed that Zeatin has an efficient and genotype independent effect on tomato explants regeneration especially when hypocotyl explant was used.
