Effects of green tea extract on expression of CLAUDIN-1 gene in gastric adenocarcinoma cell category

Abstract:

Gastric cancer, especially (carcinoma type) is one of the most prevalent disease which is categorized as the forth widespread cancer and the second cause of death related to the cancer in the world. The disease is appeared when gastric mucosal cell is cancerous because of the effect of mutation on controlling genes or as the result of helicobacter pylori infection.Based on the known effects of herbal medicine on the prevention and treatment of the disease and according to the antioxidant properties of the green tea, that controls cancerous cells’ growth, green tea is considered to study in the present work.Studying the effects of CLAUDIN-1 gene on the cancers is very important that affects cancerous cells’ invasion and or mobility.Material and Methods: AGS cells are cultivated in DMEM medium supplemented with 10% FBS, 5%CO2 at 37 ₌C of incubator with 85% of humidity. The samples are treated at 800, 1200, and 2000 μg/ml concentration of green tea extract for 72 hours, then RNA extraction and CDNA synthesis is done by the kit. Finally, expression of CLAUDIN-1 gene is done by real time PCR specific primer and GAPDH gene is used as an internal control gene and the analyzed data was calculated by 2ΔΔct .Then t-test is used for checking significance level. Data analysis show that by treating green tea extract for 72 hours at concentration of 800μg/ml, expression of Claudin-1 gene dropped 35.23 times ,and the result of the process done at concentration of 1200 and 2000 reports as 36.37 and 2.03,respectively. Therefore, based on the findings it is represented that green tea extract cause decrease in the expression of CLAUDIN-1gene as carcinogen.
