Abstract:

Background: to development and validation a simple and rapid method to show toxicological level of gamma amino butyric acid derivatives, pregabalin, vigabatrin, gabapentin in human plasma.Method: A new micellar electrokinetic capillary electrophoretic method was introduced for the determination of pregabalin. In addition a simple indirect UV detection has been utilized for detection purposes, where mesalazin was added as a probe into the background electrolyte (BGE) owning to its high molar absorptivity and similar structure to pregabalin (presence of amine and carboxylic acid moieties). In order to conduct the CE analysis, a BGE containing mesalazin (10mM), CTAB (1mM) and trisodium citrate (4%v/v, 0.1mol L-1) with pH value of 12.0 was utilized and samples were introduced to the capillary using a hydrodynamic injection at 50mbar for 25s and also applied electrokinetic injection at -10kv for 150s while pH of the sample solution adjusted in 11 for improvement at repeatability.Results: method validation performed for aqueous solution according to the FDA guideline and showed good linearity range (R2> 0.999) for 1.5-100 ppm, precision (%RSD < 10) for both migration time and peaks area. LOD and LOQ were quantified 0.8 and 2.6ppm respectively in aqueous solution with apply on-line sample stacking and hydrodynamic injection. Quantification of toxic level of the drug in plasma sample at 50ppm concentration has been done in electrokinetic injection mode (FASI).Conclusion: indirect UV detection and simultaneous applied MEKC-FASS technique at online manner in the capillary electrophoresis is a simple and rapid and cost effective for determination and quantification of pregabalin in plasma sample at toxic level.
