Relationship between PON 3 gene L 192 M polymorphism in women with idiopathic infertility in population of West Azerbaijan province

Abstract:

Background and Aim : Oxidative stress is one of the major factors in the pathophysiology ofendometriosis patients. It is located on the long arm of chromosome 7 and contains 3 genes, about 1% inthe amino acid sequence and 5% in the nucleotide sequence. This highly reactive gene promotes the growthand adhesion of endometrial cells in the perioral cavities leading to endometriosis and infertility.Methods : In this study, the population consisted of 5 infertile individuals from West Azarbaijan provinceand 5 healthy individuals as control group. Blood samples were taken from all subjects and DNA extractionwas performed by salt ethanol deposition method. PCR was used and RFLP technique was used for cuttingand sequencing of the gene. Finally, the data were edited by SPSS V.24 software.Results : The mean age of the patients was 49 ± 1.7 and 51 1 1.9 for the control group and 62% for the Aallele and 38% for the dominant homozygous (GG) genotype, respectively. There was a significantcorrelation with disease incidence (OR = 19.28 95% -CL = 2/24/27/98). Also the frequency of homozygousrecessive (AA) and heterozygous (AG) genotypes were equal to 22% and 35% had no association withdisease incidence (OR = 66.7, 95% -CL = 69.8-69.02, P = 0.009). Also, those with family marriages had a5 percent higher incidence of the disease than other people.Conclusion : Based on the results, the individuals with dominant homozygous genotype had a significantrelationship with the incidence of the disease, which can be used as one of the pre-clinical factors
