Abstract:

Glioblastoma (GBM), is the most aggressive cancer that begins within the brain. GBM has multifactorial pathogenesis, including genetic factors, epigenetic modification, and environmental elements. GBM occurs in both children and adult individuals in a very different pattern, but they can have a genetic etiology. Therefore, there are many techniques like NGS that can be used for identifying important genes in glioblastoma pathogenesis. Since, due to genetics elements roles in many diseases such as glioblastoma pathogenesis, they can be used as a biomarker in the diagnosis of many diseases especially glioblastoma in an early stage. Therefore, TNKS2 and EGFR are two most important genes, identified through NGS analysis, that play a role in glioblastoma pathogenesis and progression. Gene’sexpression profiling by array of children and adult patients with glioblastoma were extracted from Geo Data sets. Genes are compared with log fc for their expression between two kinds of patients andsorted by their p-value. Next genes with most changes in their expression were selected in each group of patients. Also the most related pathway, location and function of these genes were identified through GeneCard data sets. TNKS2 has hyperexpression in children that play a role in telomere extension by telomerase and signaling by Wnt. EGFR has overexpression in adult patients and play a role in signaling to RAS and cellular apoptosis pathway. All of these improve TNKS2 and EGFR roles in tumorigenesis of glioblastoma. Through identifying of critical genes in any disease like glioblastoma byNGS analysis, we can achieve to important biomarkers for early diagnosis. Therefore TNKS2 and EGFR can be used as diagnosis biomarker in children and adult patients of glioblastoma through NGSanalysis.
