Abstract:

Introduction: miRNAs are approximately 22-nucleotide, single-stranded RNA molecules. It is now predictedthat over 50% of human mRNAs could be regulated level by miRNAs. These miRNAs are known to controlprocesses including development, apoptosis, proliferation, differentiation, apoptose, angiogenese and immuneresponse. It has been shown that genetic factors are significant in the risk of stroke. This study was aimed todetermine the association between miR-499 (rs3746444 A/G) and miR-608 (rs4919510 C/G) variants with strokein an Iranian population. Methods: In this case-control study 206 patients with all kinds of stroke and 210 subjects without any strokehistory as the control group recruited. Levels of clinical data including hypertension, diabetes, ischemic heartattack, low density lipoprotein (LDL), high density lipoprotein (HDL), triglyceride and total cholesterol weredetermined. All kinds of strokes in cases were diagnosed by computed tomography (CT) scans and magneticresonance imaging (MRI). Polymorphisms were assessed by using polymerase chain reaction-restrictionfragment length polymorphism (PCR-RFLP) or ARMS-PCR methods. Statistical analyses were performed usingSPSS software and P-value < 0.05 was considered significant. Results: Following statistical analysis significant association was found among miR-499 (rs3746444 A/G)genetic variations and stroke in the Iranian population studied (P=0.021).Conclusion: miR-499 (rs3746444 A/G) could be considered as stroke risk factor.
