Abstract:

Background & objective: Breast cancer is considered as the most prevalent type of cancer among the women with the highest mortality rate. The purpose of the study was to examine the expression changes of STAT۳ and AP-۱ genes in MCF-۷ breast adenocarcinoma cells treated with cyclophosphamide and methotrexate using RealTime PCR quantitative method.Materials & Methods: These two drugs were prepared in various concentrations (۱, ۵ and ۱۰ micrograms / ml) and (۵, ۱۰ and ۲۵ micrograms / ml), respectively. MCF-۷ cancer cells were treated with chemotherapy drugs in various groups and times after cell passage. RNA extraction and cDNA synthesis were done and STAT۳ and AP-۱ gene expression was evaluated using RealTime PCR.Results: According to the results, both genes indicated significant reductions in various times and concentrations.Discussion & Conclusion: Based on the results obtained, MCF-۷ cell line treatment with ۲ identified drugs of STAT۳ and AP-۱ genes can be reduced in a dose and time dependent manner. This reduction can be a proof for treatment of breast cancer.
