Abstract:

In this study the effect of Archaeosomal Paclitaxel drug on Breast Cancer cell linesMCF-۷ cells was evaluated. The experiments were conducted to assess thecytotoxicity of ۷ Paclitaxel in free form and compare it with Archaeosomal form ofthe drug in vitro. To check the size of Archaeosomes made, Zeta sizer device(DLS) was used. The average size of Archaeosom Nanoparticles containingPaclitaxel drugs ۴۲۵ nm and surface charge -۱۴, the average size of controlArchaeosom Nanoparticles without drug ۴۵۸.۲ nm and surface charge -۱۳ werereported. . In both formulations zeta potential was negative. Previous studies haveshown negatively charged liposomes have high physical stability, small particlesize and high percentage of trapping the drug in the comparison with positivelycharged. Nano-Archaeosom containing Paclitaxel drug has higher physical stabilitycompared with control Nano-Archaeosom. The effect of Archaeosomal drugcontaining Paclitaxel and free drug were examined by MTT test. Tests showed thekilling effect of Nano-Archaeosome containing Paclitaxel formulation Tumor cellsis more than free formulation. In the other word, Nano-Archaeosome containingPaclitaxel is more effective than free Paclitaxel.
