Abstract:
Alzheimer's disease (AD) is a common neurological disorder characterized by the accumulationof beta amyloid (Aβ) plaques in the brain. These plaques are the main component of neuronalplaques in AD. The accumulation of Aβ is caused by the activity of an enzyme called betasecretase(BACE-۱). Nanoparticles, such as gold nanoparticles (AuNP), may play a critical rolein the secondary structure and function of BACE-۱. The interaction between AuNP and BACE-۱was investigated through ۱۸ experiments. The ratio of nanoparticle to enzyme, the type ofenzyme, and the reaction time of enzyme with nanoparticle were analyzed using Design Expertsoftware. It is important to note that nanoparticles can have toxic effects. The toxicity ofnanoparticles, including AuNP, can lead to side effects such as bad breathing, skin itching, handtremors, and memory loss. Further investigation is required to understand the potentialchallenges associated with the use of nanoparticles.The experiments conducted using DesignExpert software revealed important findings. The results showed a break point for the lowestenzyme activity, which occurred at a reaction time of ۹۰ minutes and an enzyme concentration of۸ μL/L. These findings suggest that reducing BACE-۱ activity may help alleviate AD symptoms,but it is important to consider potential interference and the possibility of increased expression ofBACE-۱. In summary, AD is characterized by the accumulation of Aβ, which is caused by theactivity of BACE-۱. Nanoparticles, such as AuNP, can influence the structure and function ofBACE-۱.
