Association Study of rs۱۰۰۹۰۱۵۴ and rs۱۶۹۱۰۵۳ With Predisposition to Prostate Adenocarcinoma in the Iranian Population

Abstract:
Prostate cancer is the second most prevalent malignancy in men and the fifth leading cause of death worldwide. It is the second most common urinary tract cancer among Iranian men. The aim of this study was to investigate the association between rs۱۰۰۹۰۱۵۴, located on the ۸q۲۴ locus, and rs۱۶۹۱۰۵۳ on chromosome ۵p۱۵.۳۱, with prostate adenocarcinoma and PSA levels. This study also aimed to identify the potential of these genetic markers as screening factors. This case-control study included ۷۹ patients with prostate adenocarcinoma aged between ۴۸ to ۸۶ years, as well as ۹۸ patients with benign prostatic hyperplasia aged between ۴۷ to ۸۱ years. The Tetra-primer ARMS-PCR method was applied to determine the genotype of each participant. In this study, no significant differences in genotypic distribution between the prostate adenocarcinoma and control groups were discovered for rs۱۰۰۹۰۱۵۴ (P-value = ۰.۶۰۸) and rs۱۶۹۱۰۵۳ (P-value = ۰.۱۰۲) polymorphisms. Moreover, for the study of the additive genetics model of the rs۱۰۰۹۰۱۵۴ polymorphism, with the TT genotype as the reference, the CC genotype with P-value = ۱ and OR {۹۵%CI} = ۰.۷۵۰, {۰.۰۳۹-۱۴.۵۷۶} and CT genotype with P-value = ۰.۳۲۴ and OR {۹۵%CI} = ۰.۵۷۷, {۰.۱۹۱-۱.۷۳۹}, were not associated. Correspondingly, for rs۱۶۹۱۰۵۳, with the CC genotype as the reference, the CT genotype with P-value = ۰.۱۷۶ and OR {۹۵%CI}= ۰.۱۹۶, {۰.۰۲۲-۱.۷۹۳}, and the TT genotype with P-value= ۰.۴۶۴, OR{۹۵%CI}= ۰.۱۲۵, {۰.۰۰۵-۳.۲۲۵}, were not associated. These findings suggest that rs۱۰۰۹۰۱۵۴ and rs۱۶۹۱۰۵۳ may not be associated with prostate cancer among Iranians. However, further research with larger sample sizes and investigation of various Iranian subpopulations are needed to confirm these results.
