Abstract:

Habbatussauda or black cumin, Nigella sativa, is an herbal ingredient used for by community as medicine. It contains tannins, thymoquinone, thymol, pinene, and p-cymene that inhibit the bacterial growth of both positive and negative grams. Streptococcus mutans is a primary bacterium that makes caries on teeth since it has a glucosyltransferase enzyme, or GFT. This enzyme could catalyse glucan synthesis in the process of tooth caries. This study determines the influence of the mouthwash with black cumin extract, Nigella sativa, in inhibiting the bacterial growth of Streptococcus mutans within the in vitro situation. This laboratory experimental research applied a post-test-only control group design. The applied bacterial activity test was well diffusion. The researchers grouped the samples into six treatment samples: the mouthwash solution with concentrations of ۳%, ۶%, ۹%, and ۱۲%; and the negative and positive control groups. The positive control group used mouthwash with ۰.۲% chlorhexidine. Each group consisted of four samples. Thus, the total samples were ۲۴. The test of the bacterial growth inhibiting power applied the well technique. The results of this study revealed that the extracted black cumin mouthwash solution with concentrations of ۳%, ۶%, ۹%, and ۱۲% influenced the Streptococcus Mutans bacterial growth inhibition. The ANOVA test result obtained a significant value or (p) less than ۰.۰۰۵, p < ۰.۰۵. The Post Hoc LSD test found the differences among the groups with the inter-group significance of p < ۰.۰۵. The usage of the extracted black cumin mouthwash, Nigella sativa, influenced the inhibiting zone of the Streptococcus mutans bacterial growth in an in vitro manner. The mouthwash with ۹% black cumin has the highest effect to inhibit the Streptococcus mutans bacterial growth.
