Glycine nitrate: a novel and green organocatalyst for synthesis of 3,4-dihydropyrimidin-2(1H)-ones under solvent-free conditions

Biginelli reaction is one of the most important multi-component reactions for the synthesis of dihydropyrimidinones. These category of heterocyclic compounds have interesting pharmacological properties such as antiviral, antibacterial, and antihypertensive effects, and used as calcium channel modulators, and as multi drug resistance reversal. Futhermore, the 2-oxo-dihydropyrimidine-5-carboxylate core unit is found in many marine natural products including batzelladine alkaloids, which have been found to be HIV-gp-120 CD4 inhibitors. In this study, several compounds with dihydropyrimidines skeletal wereprepared by reaction of aldehyde, urea and β-keto ester. A highly efficient and improved synthetic methodology for the preparation of dihydropyrimidinone derivatives using ionic liquid under solvent-free conditions was used. This methodology offers several advantages such as high yields, clean process, simple experimental work-up procedure and low loading of catalyst.
