Abstract:
IntroductionClassic in vitro 2-dimensional (2D) cancer cell cultures are hypersensitive to anti-cancer drugs. So, development of three dimensional (3D) cultures to better replicate natural physiology and better predict drug efficacy, is necessary. Objectives Here we introduce a novel 3D model of gastric cancer using decellularized gastric tissue. MethodsRat gastric tissue after heart perfusion by normal saline and 1% SDS was extracted. Gastric tissue placed in SDS 1% for 3 hours and washed with PBS. MKN-45 Cells were injected into decellularized tissue specimen and cultured for 7 days. Decellularized tissue before and after cell injection examined with H&E staining and SEM. ResultsThe results showed that gastric tissue can be easily decellularized with SDS. MKN-45 cells were growth in decellularized gastric tissue easily and form tumor like structure. ConclusionDecellularized gastric tissue can be used as a proper 3D matrix for gastric cancer research
