Abstract:

Among species of roses, Rosa damascena Mill is with very high important economic and commercial value. These species are cultivated for their essential oil, water flowers which are widely used in the medicine, food and cosmetics industries. This study was carried out as a step toward asexual propagation in four genotypes. Apical and axillary buds from adult elite trees of four genotypes were collected in different seasons. Immersion buds in %0.1 mercuric chloride solution in different time 6 and 4 minutes for Illam and Esfahan and as well as %10 sodium hypochlorite in 10 minutes and 0.1 % mercuric chloride solution in time 2 minutes for Ardabil and Semnan were the best treatments among surface sterilization tests respectively. The best season in order to collect samples was summer. These explants were cultured on a modified MS medium containing cytokinines (BAP, Kinetin, TDZ) in (3, 2, 0.05 mg l- 1) and auxines (IBA) in (0.01-0.5 mgl-1). Four genotypes were rooted in 1/4 MS with 10 mol-1 auxines hormone IBA and NAA. These plants were established in the greenhouse conditions (Ardabil 1 with the 45% survival, Esfahan 1 with the 50% survival, Illam 1 with the 69% survival and Semnan 1 with the 66% survival genotypes) successfully.
